It may be used as monotherapy or in combination with other antihypertensive agents such as ACE inhibitors, angiotensin receptor blockers, or diuretics.
Levamlodipine selectively inhibits the transmembrane influx of calcium ions into vascular smooth muscle and cardiac muscle by blocking L-type calcium channels. This inhibition leads to relaxation of vascular smooth muscle, resulting in peripheral arterial vasodilation and a subsequent reduction in blood pressure.
As the S-enantiomer of amlodipine, levamlodipine has higher receptor affinity and greater pharmacological potency, allowing effective antihypertensive action at lower doses. It has minimal effects on heart rate and myocardial contractility, making it suitable for long-term use in a wide range of patients.
Levamlodipine is slowly absorbed following oral administration and provides a gradual onset and sustained duration of action, supporting once-daily dosing.
Recommended starting dose: 1.25 mg once daily
