It is indicated for Irritable Bowel Syndrome with Constipation (IBS-C), Chronic Idiopathic Constipation (CIC), and Opioid-Induced Constipation (OIC) in adults with chronic non-cancer pain. Lubistone 24 mcg selectively activates ClC-2 chloride channels on the apical membrane of the intestinal epithelium. This action triggers chloride ion efflux into the intestinal lumen, followed by sodium ions through a paracellular pathway to maintain electrical neutrality. Water follows these ions, increasing intestinal fluid secretion. The result is enhanced intestinal motility, improved stool passage, and relief of constipation symptoms.
In addition to its secretory effect, Lubistone may help restore mucosal barrier function by supporting tight junction proteins in the intestinal lining. This targeted luminal activity ensures minimal systemic absorption and reduced risk of systemic side effects.
Lubistone 24 mcg is indicated for:
Irritable Bowel Syndrome with Constipation (IBS-C) in adults
Chronic Idiopathic Constipation (CIC) in adults
Opioid-Induced Constipation (OIC) in adult patients with chronic, non-cancer pain
IBS-C: 8 mcg orally twice daily with food and water. For severe hepatic impairment, start with 8 mcg once daily, increasing to twice daily if tolerated.
CIC & OIC: 24 mcg orally twice daily with food and water. In moderate hepatic impairment, start with 16 mcg twice daily; in severe impairment, start with 8 mcg twice daily.
Lubistone should be taken with meals to improve tolerability and reduce nausea.
Diphenylheptane opioids may reduce Lubistone’s efficacy in a dose-dependent manner.
Known or suspected mechanical gastrointestinal obstruction
Diarrhea
Abdominal bloating or fullness
Nausea
Stomach pain
Abdominal swelling
Dyspnea
Patients with severe diarrhea should avoid Lubistone
Evaluate for mechanical bowel obstruction before initiation
Monitor infants for diarrhea if administered during breastfeeding
Nausea may be reduced by taking the drug with food
Symptoms may include: nausea, diarrhea, vomiting, dizziness, headache, abdominal pain, flushing, dyspnea, pallor, chest discomfort, dry mouth, hyperhidrosis, syncope, and anorexia. Supportive care is recommended in case of overdose.
